DETECTR (CRISPR-Cas12a Diagnostics)

Demonstration that Cas12a, once activated by binding its double-stranded DNA target, indiscriminately cleaves nearby single-stranded DNA reporters -- the DNA-targeting analogue of Cas13a's RNA collateral-cleavage activity (see the linked SHERLOCK node) -- repurposed into a rapid nucleic-acid diagnostic. Chen, Ma, Harrington, Da Costa, Tian, Palefsky & Doudna (Jennifer Doudna's lab), "CRISPR-Cas12a target binding unleashes indiscriminate single-stranded DNase activity" (Science 360, 436-439, 2018-02-15, OpenAlex W2787024379, 4,560 citations). Commercialized as the DETECTR platform by Mammoth Biosciences; another genuine "second wave" diagnostic translation of the CRISPR-Cas mechanism family (see the linked precursor node), independent of, but published within months of, the Zhang-lab SHERLOCK platform above.
